This new method may accelerate research and clinical applications for stem cell corneal transplantation. via ScienceDaily: Top Health News: Nov. 27, 2013 — Researchers in the Cedars-Sinai Regenerative Medicine Institute have designed and tested a novel, minute-long procedure to prepare human amniotic membrane for use as a scaffold for specialized stem cells that may be used to treat some corneal diseases. This membrane serves as a foundation that supports the growth of stem cells in order to graft them onto the cornea.This new method, explained in a paper published this month in the journal PLOS ONE, may accelerate research and clinical applications for stem cell corneal transplantation.Corneal blindness affects more than 8 million people worldwide. Among other causes, corneal blindness can be the outcome of corneal stem cell deficiency, a disease usually resulting from genetic defects or injury to the eye — such as burns, infection or chronic inflammation — that can lead to vision loss. A feasible treatment to rectify vision loss for such patients is corneal stem cell transplantation, either as a biopsy from another eye or by transplanting cultured stem cells, although this promising approach is not yet fully standardized.An approved biological foundation for cultured stem cells is the human amniotic membrane, a thin but sturdy film that separates the fetus from the placenta. For the best growth of stem cells, amniotic cells need to be removed by chemical agents. A teragram is equal to 1 million tons.”It is now on par with the methane being released from the arctic tundra, which is considered to be one of the major sources of methane in the Northern Hemisphere,” said Natalia Shakhova, one of the paper’s lead authors and a scientist at the University of Alaska Fairbanks. “Increased methane releases in this area are a possible new climate-change-driven factor that will strengthen over time.”Methane is a greenhouse gas more than 30 times more potent than carbon dioxide. Climate change has silver lining for grizzly bears Global warming and forest disturbances may have a silver lining for threatened species of grizzly bears in Alberta, Canada. via ScienceDaily: Top Science News: Oct. 28, 2013 — Global warming and forest disturbances may have a silver lining for threatened species of grizzly bears in Alberta, Canada.In a 10-year study that monitored 112 bears in Alberta’s Rocky Mountain region, University of Alberta biologist Scott Nielsen and his colleagues found that warmer temperatures and easier access to food associated with forest disturbances helped the grizzlies to build more body fat, known to increase the chances of successful reproduction for mothers.The resulting ‘silver spoon effect’ shows that bears born into these favourable conditions have a head-start in life, said Nielsen, an assistant professor in the U of A Department of Renewable Resources.”Understanding variations in body size helps us understand what limits grizzly populations,” Nielsen said. “We get clues about the environments that most suit grizzlies by examining basic health measures such as body size. A simple rule is, the fatter the bear, the better. Certain environments promote fatter bears.The findings, published in BMC Ecology, may help influence forest harvest designs to enhance habitat for the Alberta grizzly, which is classed by the Alberta government as a threatened species. Currently there are only about 750 of the bears in the province, half of them adults.In years when warmer temperatures and less late winter snow brought on earlier spring conditions, the body size of bears as adults was larger. Together with rhinoviruses A and B, the recently discovered virus is responsible for millions of illnesses yearly at an estimated annual cost of more than $40 billion in the United States alone.The work is important because it sculpts a highly detailed structural model of the virus, showing that the protein shell of the virus is distinct from those of other strains of cold viruses.“The question we sought to answer was how is it different and what can we do about it? We found it is indeed quite different,” says Palmenberg, noting that the new structure “explains most of the previous failures of drug trials against rhinovirus.”The A and B families of cold virus, including their three-dimensional structures, have long been known to science as they can easily be grown and studied in the lab. Rhinovirus C, on the other hand, resists culturing and escaped notice entirely until 2006 when “gene chips” and advanced gene sequencing revealed the virus had long been lurking in human cells alongside the more observable A and B virus strains.The new cold virus model was built “in silico,” drawing on advanced bioinformatics and the genetic sequences of 500 rhinovirus C genomes, which provided the three-dimensional coordinates of the viral capsid.“It’s a very high-resolution model,” notes Palmenberg, whose group along with a team from the University of Maryland was the first to map the genomes for all known common cold virus strains in 2009. “We can see that it fits the data.”With a structure in hand, the likelihood that drugs can be designed to effectively thwart colds may be in the offing. Drugs that work well against the A and B strains of cold virus have been developed and advanced to clinical trials. However, their efficacy was blunted because they were built to take advantage of the surface features of the better known strains, whose structures were resolved years ago through X-ray crystallography, a well-established technique for obtaining the structures of critical molecules.Because all three cold virus strains all contribute to the common cold, drug candidates failed as the surface features that permit rhinovirus C to dock with host cells and evade the immune system were unknown and different from those of rhinovirus A and B.Based on the new structure, “we predict you’ll have to make a C-specific drug,” explains Holly A.
